自杂交瘤技术建立以来,抗各类肿瘤的单克隆抗体(McAb)陆续建立。对颅内最常见的恶性肿瘤——人脑胶质瘤McAb除国外有少量报道外,国内尚未见报道。我们采用本研究室建立的我国第一株人脑胶质瘤细胞系SHG 44为抗原,制备了抗人脑胶质瘤McAb。应用SHG-44完整细胞免疫BaLb/C小鼠,经加强免疫后,第4天取脾脏细胞与小鼠骨髓瘤细胞系NS-1融合,96孔培养板232个孔中,93孔出现杂交瘤细胞克隆。经用酶联免疫吸附
Since the establishment of hybridoma technology, monoclonal antibodies (McAbs) against various types of tumors have been established. The most common malignant tumor in the brain, the human brain glioma McAb, has not been reported in China except for a few reports abroad. We used the first human glioma cell line SHG 44 established in our laboratory as an antigen to prepare anti-human glioma McAb. The BALb/C mice were immunized with SHG-44 whole cells. After the booster immunization, the spleen cells were fused with the mouse myeloma cell line NS-1 on the 4th day. Hybridomas appeared in 232 wells of the 96-well culture plate in 93 wells.
