目的 :探讨糖尿病肾病系膜细胞功能改变及意义。方法 :采用STZ复制糖尿病模型 ,取肾进行系膜细胞培养 ,3 H -TdR法测定细胞增殖 ,用fluro - 3探针经共聚焦显微镜检测 [Ca2 + ]i 变化 ,ELISA法测定纤维连接蛋白 (FN)分泌量。结果 :糖尿病大鼠系膜细胞3 H -TdR掺入率和FN分泌量较正常大鼠增加 ,基础 [Ca2 + ]i 两组无差异 ,但糖尿病系膜细胞 [Ca2 + ]i 对血管紧张素II反应明显减弱。结论 :糖尿病系膜细胞增殖和分泌FN增加参与了糖尿病肾病细胞外基质的积聚 ,其 [Ca2 + ]i对AngII反应减弱可能参与了糖尿病肾病早期高滤过
Objective: To investigate the changes of mesangial cell function in diabetic nephropathy and its significance. Methods: The model of STZ - induced diabetes mellitus was established. The mesangial cells were harvested from the kidney. Cell proliferation was measured by 3 H - TdR method. The changes of [Ca2 +] i were detected by confocal microscopy with fluro - 3 probe. The expression of fibronectin FN) secretion. Results: Compared with normal rats, the incorporation of 3H-TdR and the secretion of FN in mesangial cells of diabetic rats were increased. There was no difference in basic [Ca2 +] i between the two groups. However, [Ca2 +] i in diabetic mesangial cells had no effect on angiotensin II reaction was significantly weakened. CONCLUSION: Increased proliferation and secretion of FN in mesangial cells of diabetic rats are involved in the accumulation of extracellular matrix in diabetic nephropathy. The decreased response of [Ca2 +] i to AngII may be involved in the early hyperfiltration of diabetic nephropathy
