目的 :探讨吸烟对精子核DNA成熟度和精子膜功能的影响。 方法 :自制吸烟机将大鼠制成被动吸烟模型 ,取其精子应用精子核吖啶橙荧光染色法和精子低渗肿胀试验 ,观察吸烟大鼠精子核DNA成熟度和精子膜功能。结果 :大鼠被动吸烟 8周后其双链DNA精子百分率为 (5 2 0± 5 6 ) % ,明显低于相应的对照组 [(6 8 5± 3 3) % ](P <0 0 1) ;精子低渗肿胀率 [(2 0 7± 4 3) % ]也较正常对照组 [(5 2 5± 3 8) % ],明显降低 (P <0 0 1)。 结论 :吸烟可致大鼠精子核DNA成熟度降低并使大鼠精子膜更脆弱 ,降低大鼠精子受精能力。
Objective: To investigate the effects of smoking on sperm DNA maturation and sperm membrane function. Methods: The rats were made into passive smoking model by self-made smoking machine. The sperm were stained with acridine orange fluorescence staining and sperm hypoosmotic swelling test to observe the sperm DNA maturation and sperm membrane function in smoking rats. RESULTS: The percentage of double-stranded DNA in passive smoking group was (5 2 0 ± 5 6)% after 8 weeks of passive smoking, which was significantly lower than that of the corresponding control group [(6 8 5 ± 3 3)%] (P 0 01 ); Sperm hypoosmotic infiltration rate [(2 0 7 ± 4 3)%] was also significantly lower than that of the control group [(52 5 ± 38)%] (P 0 01). Conclusion: Smoking can reduce the maturation of sperm DNA in rats and make the sperm membrane more fragile and reduce the sperm fertilization ability in rats.
