目的:探讨白细胞介素-4(IL-4)和白细胞介素-13(IL-13)基因多态性对哮喘患儿Ig E表达的影响。方法:采用聚合酶链式反应(PCR)检测173例哮喘患儿及56例健康儿童的IL-4和IL-13基因限制性长度片段,及不同基因型患儿Ig E的表达水平。结果:哮喘患儿IL-4基因-589位点基因多态性和等位基因与健康对照儿童比较差异无统计学意义(P>0.05)。哮喘患儿IL-13基因+1923位点基因多态性和等位基因与健康对照儿童比较差异有统计学意义(P<0.01)。IL-4基因-589和IL-13基因+1923位点T等位基因患儿Ig E水平显著高于C等位基因患儿(P<0.01)。结论:哮喘患儿IL-13基因多态性与健康儿童不同,IL-4基因-589位点和IL-13基因+1923T等位基因是导致哮喘患儿Ig E水平升高的可能原因。
Objective: To investigate the effect of interleukin-4 (IL-4) and interleukin-13 (IL-13) gene polymorphisms on IgE expression in asthmatic children. Methods: The restriction fragment length polymorphisms of IL-4 and IL-13 genes in 173 asthmatic children and 56 healthy children were detected by polymerase chain reaction (PCR) and the Ig E expression in children with different genotypes. Results: There was no significant difference in gene polymorphism at -589 locus and allele between children with asthma and children with asthma (P> 0.05). The gene polymorphism at +1923 locus and the allele of IL-13 gene in children with asthma were significantly different from those in healthy controls (P <0.01). The level of IgE in children with IL-4 gene -589 and IL-13 gene +1923 locus was significantly higher than that in children with C allele (P <0.01). CONCLUSIONS: IL-13 gene polymorphism in children with asthma is different from healthy children. IL-4 gene -589 locus and IL-13 gene + 1923T allele are the possible causes of elevated IgE in asthmatic children.
