目的:采用响应面分析法优选丹参多糖的纤维素酶提取工艺。方法:在单因素试验基础上,利用Design-Expert8.0.5软件釆用三因素三水平响应面分析法,以加酶量、酶解温度、酶解时间为考察因素,多糖提取率为响应值,进行回归分析,优选丹参多糖提取工艺条件。结果:最佳提取工艺为:加酶量0.5%,酶解温度65℃,提取时间120 min,模型相关系数R2=0.979 5,拟合度较好。在该条件下,丹参多糖的提取率2.59 mg·g~(-1),与理论提取率(2.51 mg·g~(-1))接近。结论:该提取工艺客观可行、稳定合理,可为工业化生产提供理论依据。
Objective: To optimize the extraction process of polysaccharide from Salvia miltiorrhiza by response surface methodology. Methods: On the basis of single factor experiment, using Design-Expert8.0.5 software, preclude the use of three factors and three levels response surface analysis method, with enzyme dosage, enzymolysis temperature and enzymolysis time as the investigation factors, polysaccharide extraction rate as the response value, Regression analysis, optimization Salvia polysaccharide extraction process conditions. Results: The optimum extraction conditions were as follows: the amount of enzyme was 0.5%, the temperature of enzymolysis was 65 ℃, the extraction time was 120 min, the model correlation coefficient was R2 = 0.979 5, the fitting degree was better. Under this condition, the extraction rate of polysaccharides from Salvia miltiorrhiza was 2.59 mg · g ~ (-1), close to the theoretical extraction rate (2.51 mg · g ~ (-1)). Conclusion: The extraction process objectively feasible, stable and reasonable, can provide a theoretical basis for industrial production.
