取田间种植的大麦品种‘花30’幼穗游离小孢子,采用荧光素二醋酸酯(FDA)染色法测定其在不同Na Cl浓度胁迫下的活力,用Image-Pro Plus软件对显微系统拍摄的照片进行分析计数,研究了大麦离体小孢子对盐胁迫的耐受性。结果表明:利用FDA染色结合软件显微拍照系统分析可以快速、准确地测定和分析游离小孢子的活力和大小。对本年度大田种植的大麦‘花30’材料,按照成熟方法游离小孢子,获得40%具有活力的小孢子,游离小孢子在含1%Na Cl提取液中至少2 h内活力保持稳定。
Microspores were isolated from field microspores by field-grown barley cultivar ’Hua’30’. Microspheres were harvested with Image-Pro Plus software using fluorescein diacetate (FDA) staining to determine their activity under different NaCl concentrations. The photos were analyzed and counted to study the salt tolerance of the microspore isolated from barley. The results showed that the activity and size of the free microspore could be quickly and accurately determined and analyzed by the FDA system software combined with the microscopic photographing system. For the barley cultivar ’Hua30’ grown in the field in this year, the microspores were detached according to the mature method to obtain 40% viable microspores. The free microspores remained stable in at least 2 h with 1% NaCl extract.
