目的探讨姜黄素对应激状态下大鼠胃溃疡的防治作用及机制。方法将40只雄性SD大鼠随机分为:正常对照组、溃疡模型组、姜黄素组、奥美拉唑组,每组10只。采用水浸-束缚方法建立大鼠胃溃疡模型,检测各组胃黏膜溃疡指数(UI)、胃液pH值和壁细胞H+,K+-ATP酶活性,观察其胃黏膜组织病理变化,采用SPSS13.0软件进行统计学分析。结果与正常对照组比较,溃疡模型组UI(35.63±3.34)明显升高,胃液pH值(1.35±0.09)明显下降,壁细胞H+,K+-ATP酶活性(9.45±0.34)mmol/(g.h)明显升高(Pa<0.01),镜下可见胃黏膜出血坏死,溃疡形成。与溃疡模型组比较,姜黄素组UI(15.13±2.42)明显下降,胃液pH值(1.81±0.23)明显上升,壁细胞H+,K+-ATP酶活性(8.13±0.59)mmol/(g.h)明显下降(Pa<0.01),镜下胃黏膜损伤程度较轻,未见溃疡形成。姜黄素组与奥美拉唑组比较无显著差异(Pa>0.05)。胃液pH值和壁细胞H+,K+-ATP酶活性水平呈明显负相关(r=-0.884 P<0.01)。结论姜黄素可有效地减轻大鼠胃溃疡模型的胃黏膜损伤,抑制壁细胞H+,K+-ATP酶活性,减少胃酸分泌,对胃溃疡有良好的防治作用。
Objective To investigate the curative effect and mechanism of curcumin on rat gastric ulcer under stress. Methods Forty male Sprague-Dawley rats were randomly divided into normal control group, ulcer model group, curcumin group and omeprazole group, with 10 rats in each group. Rat gastric ulcer model was established by water immersion-binding method. The gastric mucosal ulcer index (UI), pH value of gastric juice, and H+, K+-ATPase activity of parietal cells were detected. Histopathological changes of gastric mucosa were observed. SPSS13.0 was used. The software performs statistical analysis. Results Compared with the normal control group, the UI in the ulcer model group was significantly higher (35.63±3.34), the gastric juice pH value (1.35±0.09) was significantly decreased, and the parietal cell H+, K+-ATPase activity was (9.45±0.34) mmol/(gh). Significantly increased (Pa <0.01), gastric mucosal hemorrhage and necrosis, ulcer formation. Compared with the ulcer model group, the UI (15.13±2.42) in the curcumin group decreased significantly, the gastric pH value (1.81±0.23) increased significantly, and the parietal cell H+, K+-ATPase activity (8.13±0.59) mmol/(gh) decreased significantly. (Pa<0.01), the degree of gastric mucosal damage under the microscope was mild, no ulcer formation. There was no significant difference between curcumin group and omeprazole group (Pa>0.05). The pH value of gastric juice was significantly negatively correlated with the H+, K+-ATPase activity of parietal cells (r=-0.884 P<0.01). Conclusion Curcumin can effectively reduce the gastric mucosal damage in rat gastric ulcer model, inhibit the activity of H+ and K+-ATPase in parietal cells, reduce gastric acid secretion, and have a good preventive and therapeutic effect on gastric ulcer.
