目的:观察凤尾草总黄酮对人骨肉瘤MG-63细胞体外迁移能力的影响,并探讨其作用机制。方法:利用乙醇萃取法制备凤尾草总黄酮,通过划痕实验、Transwell实验观察不同浓度凤尾草总黄酮对同样条件培养下骨肉瘤MG-63细胞迁移能力的影响,并采用Western blot方法检测不同浓度凤尾草总黄酮对MG-63细胞内白介素6(IL-6)蛋白表达水平的影响。结果:同未加药物对照组相比,经高浓度凤尾草总黄酮(5与10 g.L-1)处理后,MG-63细胞的划痕愈合能力明显减弱,穿膜细胞的数量显著减少;同时,细胞内IL-6蛋白的表达水平明显降低,并与凤尾草总黄酮的浓度呈负相关性。结论:凤尾草总黄酮抑制了骨肉瘤MG-63细胞的迁移能力,其作用与抑制MG-63细胞内的IL-6蛋白表达水平有关。
Objective: To observe the effect of Pteris total flavonoids on the in vitro migration of human osteosarcoma MG-63 cells and to explore its mechanism. Methods: The total flavonoids of Pteris sibiricum were prepared by ethanol extraction. The effects of different concentrations of Pteris flavus on the migration of osteosarcoma MG-63 cells were observed by scratch test and Transwell assay. Western blot was used to detect Effect of Different Concentrations of Total Flavonoids from Pteridium aquilinum on the Expression of Interleukin 6 (IL-6) in MG-63 Cells. Results: Scratch healing ability of MG-63 cells was obviously weakened and the number of transmembrane cells was significantly decreased after treated with high concentration of Pteris total flavonoids (5 and 10 gL-1) At the same time, the expression of IL-6 in cells was significantly decreased, which was negatively correlated with the concentration of total flavonoids in Pteris vittata. CONCLUSION: Total flavonoids of Psidium sp. Inhibits the migration of osteosarcoma MG-63 cells, which is related to the inhibition of IL-6 protein expression in MG-63 cells.
