目的:探讨18-β甘草次酸(glycyrrhetinic acid,GA)对大鼠肾脏的影响。方法:120只Wistar大鼠随机平均分成正常组及低、中、高剂量18-β甘草次酸组(25,50,100 mg·kg~(-1)),共4组,于甘草次酸ig干预后第6,14,22,30周及停药4周后心脏采血,检测血肌酐(SCr),尿素氮(BUN)含量,HE染色进行肾脏组织病理学观察。结果:随着干预进行直至停药4周时,低、中、高剂量甘草组SCr,BUN与正常组相比均无明显差异。血钠、氯离子均较正常组升高,但停药4周时血钠、氯离子较之前各时间点降低,并接近正常组。血钾离子均低于正常组,停药4周时血钾离子较之前各时间点升高并接近正常组。上述各指标同一时间点低、中、高剂量甘草组两两之间无统计差异。HE染色见各时间点肾脏病理形态与正常组相比未见异常改变。结论:18-β甘草次酸对大鼠血尿素氮、肌酐值及肾脏病理形态无明显影响。可引起血钠、氯离子轻度升高,血钾离子降低,停药一定时间后上述影响可消除。
Objective: To investigate the effect of 18-β glycyrrhetinic acid (GA) on rat kidney. Methods: One hundred and twenty Wistar rats were randomly divided into normal group and low, medium and high doses of 18-β glycyrrhetinic acid group (25, 50, 100 mg · kg -1) At the 6th, 14th, 22th, 30th and the 4th week after the withdrawal, heart blood was taken for measurement of serum creatinine (SCr) and blood urea nitrogen (BUN). HE staining was used to observe the histopathology of the kidneys. Results: There was no significant difference in SCr, BUN between the low, medium and high dose licorice groups as compared with the normal group as the intervention stopped until 4 weeks. Serum sodium and chloride were higher than those in the normal group, but serum sodium and chloride were lower at 4 weeks after drug withdrawal and were close to the normal group. Blood potassium levels were lower than the normal group, withdrawal 4 weeks when blood potassium levels increased compared with the previous time points and close to the normal group. The above indicators at the same time point low, medium and high doses of licorice group no statistically significant differences between the two. HE staining at each time point showed no change in the pathological morphology of the kidney compared with the normal group. Conclusion: 18-β-glycyrrhetinic acid has no significant effect on blood urea nitrogen, creatinine and renal pathological changes in rats. Can cause blood sodium, chloride ion mildly elevated serum potassium decreased, withdrawal of certain time after the above effects can be eliminated.
