目的:优选ZTC 1+1-Ⅱ型澄清剂处理黄瑞香叶水提液的工艺条件。方法:以祖师麻甲素保留率和固形物去除率为指标,通过单因素试验考察药液质量浓度、温度、搅拌速度、水浴保温时间、澄清剂组份加入顺序和加入量对澄清效果的影响。采用HPLC测定祖师麻甲素含量,流动相甲醇-0.5%乙酸(25∶75),检测波长327 nm。结果:最佳澄清工艺为药液生药质量浓度0.1 g·mL-1,温度80℃,加入B组份溶液后加入A组份溶液,加入量分别为6%,3%,搅拌速度100 r·min-1,水浴保温时间10 min;祖师麻甲素保留率74.29%,固形物去除率38.80%。结论:ZTC 1+1-Ⅱ型澄清剂处理黄瑞香叶提取液的澄清效果良好,优选的澄清工艺稳定可行。
Objective: To optimize the process conditions of ZTC 1 + 1- Methods: According to the retention rate of A and Z, the removal rate of solids was used as an index to study the influence of the concentration, temperature, stirring speed, holding time of the water bath, order and quantity of clarification agent on the clarification effect by single factor test . The HPLC method was used to determine the content of ephedrine, the mobile phase was methanol-0.5% acetic acid (25:75) and the detection wavelength was 327 nm. Results: The optimum clarification process was that the concentration of crude drug was 0.1 g · mL-1 and the temperature was 80 ℃. After the solution of component B was added, the solution of component A was added at 6%, 3% and 100 r · min-1, water bath holding time 10 min; founder Tiamene 74.29% retention rate, solids removal rate of 38.80%. Conclusion: The clarification effect of ZTC 1 + 1-Ⅱ clarifying agent on the leaf extract of Huangrui Xiang is good, and the preferred clarification process is stable and feasible.
