目的:利用高效液相色谱法对西瓜藤中豆甾醇和β-谷甾醇的含量进行测定。方法:取粉碎的西瓜藤10g,本实验采取超声提取法,选择溶剂为100mL甲醇,超声处理1h;色谱柱为Hypersil ODS2(4.6mm×250mm,5μm);流动相为甲醇-水(97∶3),流速:1mL/min;紫外检测波长:208nm;柱温:25℃。结果:测得豆甾醇在0.49~4.90μg范围内线性关系良好,回归方程Y=230.54X+53.255,(R2=0.994);β-谷甾醇在0.51~5.10μg范围内线性关系良好,回归方程Y=153.39X+54.198,(R2=0.993)。结论:高效液相色谱法快速简便,准确度高,重现性好,可作为西瓜藤中豆甾醇和β-谷甾醇的含量测定方法。
OBJECTIVE: To determine the contents of stigmasterol and β-sitosterol in watermelon rattan by high performance liquid chromatography. Methods: Ultrasonic extraction was used in this experiment. The solvent was 100 mL of methanol and sonicated for 1 h. The column was Hypersil ODS2 (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol-water (97: 3 ), Flow rate: 1 mL / min; UV detection wavelength: 208 nm; column temperature: 25 ° C. Results: The calibration curve was linear in the range of 0.49 ~ 4.90μg. The regression equation was Y = 230.54X + 53.255 (R2 = 0.994). The linear relationship of β-sitosterol in the range of 0.51 ~ 5.10μg was good. = 153.39X + 54.198, (R2 = 0.993). Conclusion: HPLC is rapid, simple, accurate and reproducible. It can be used as a method for the determination of stigmasterol and β-sitosterol in watermelon.
