目的观察秋水仙碱对家兔无菌性心包炎模型快速房性心律失常(RAA)发生情况的影响,并探讨其可能机制。方法将36只健康成年雄性家兔随机分为假手术组、心包炎组和秋水仙碱组,均n=12。三组均给予短阵高频刺激,心包炎组、秋水仙碱组建立无菌心包炎模型。记录体表心电图(ECG),观察房性心律失常发生情况。秋水仙碱干预结束后按分组取右心耳组织行HE染色及荧光定量PCR分析。结果与心包炎组相比,术后第5日秋水仙碱组RAA发生率下降45.4%,炎症细胞浸润率下降54.5%。心包炎组kir3.4 mRNA表达水平明显高于秋水仙碱组(P<0.05)。结论秋水仙碱能减少家兔无菌性心包炎模型RAA的发生,其可能机制是减少心房肌组织炎症反应、降低kir3.4 mRNA表达水平。
Objective To observe the effect of colchicine on the occurrence of atrial tachyarrhythmia (RAA) in rabbits with aseptic pericarditis and to explore its possible mechanism. Methods Thirty - six healthy adult male rabbits were randomly divided into sham operation group, pericarditis group and colchicine group, all with n = 12. Three groups were given short-term high-frequency stimulation, pericarditis group, colchicine group to establish aseptic pericarditis model. Record body surface electrocardiogram (ECG) to observe the occurrence of atrial arrhythmia. Colchicine after the intervention by grouping the right atrial appendage HE staining and quantitative PCR analysis. Results Compared with pericarditis group, the incidence of RAA decreased 45.4% and the inflammatory cell infiltration rate decreased 54.5% after colchicine treatment on the 5th postoperative day. The expression of kir3.4 mRNA in pericarditis group was significantly higher than that in colchicine group (P <0.05). Conclusion Colchicine can reduce the incidence of RAA in aseptic pericarditis model of rabbits. The possible mechanism is to reduce the atrial fibrillation and reduce the expression of kir3.4 mRNA.
