目的 探讨凋亡相关基因产物Fas和FasL蛋白在肾癌发生中的作用以及与转移、预后的关系。 方法 采用免疫组织化学方法对 46例肾癌组织和 15例正常肾组织Fas和FasL蛋白的表达进行检测。 结果 肾癌组织Fas表达率为 18.14% ,低于正常肾组织 (4 6 .15 % ,P <0 .0 5 ) ,随肾癌分级的增加 ,表达强度下降。肾癌组织FasL蛋白表达率为 70 .2 6 % ,高于正常肾组织 (10 .32 % ,P <0 .0 5 ) ,随肾癌分级的增加 ,表达强度增高。正常肾组织Fas与FasL的表达有相关性 (r =0 .6 89,P <0 .0 5 ) ,肾癌组织无相关性 (r =0 .143 ,P >0 .0 5 ) ,有淋巴结转移组FasL(89.42 % )与无淋巴结转移组 (6 0 .39% )比较 ,差别有显著性意义 (P <0 .0 1)。生存率 >5年组Fas(2 5 .39% )和FasL(6 1.2 6 % )与生存率 <5年组Fas(15 .2 4% )和FasL(85 .35 % )的差别均有显著性意义 (P均 <0 .0 5 )。结论 Fas和FasL蛋白相互作用失衡在肾癌的发生、发展中起重要作用 ,表达情况与肾癌病理分级及转移、预后有一定关系。
Objective To investigate the role of apoptosis-related gene products Fas and FasL in the development of renal cell carcinoma and its relationship with metastasis and prognosis. Methods Immunohistochemistry was used to detect the expression of Fas and FasL protein in 46 cases of renal cell carcinoma and 15 cases of normal renal tissues. Results The positive expression rate of Fas in renal cell carcinoma was 18.14%, which was lower than that in normal renal tissues (46.5%, P <0.05). The expression of Fas was decreased with the increase of renal cell carcinoma. The expression of FasL protein in renal cell carcinoma was 70.26%, which was higher than that in normal renal tissues (10.32%, P <0.05). The expression of FasL protein increased with the increase of renal cell carcinoma. The expression of Fas and FasL in normal renal tissue was correlated (r = 0.96, P <0.05), but not in renal cancer (r = 0.143, P> 0.05) The difference between FasL (89.42%) and non-lymph node metastasis (60.39%) in metastasis group was significant (P <0.01). The difference of Fas (15.24%) and FasL (85.35%) between survivals> 5 years and FasL (6 1.2 6%) and survival rates <5 years were significant Sexual significance (all P <0.05). Conclusion The imbalance of the interaction between Fas and FasL plays an important role in the occurrence and development of renal cell carcinoma. The expression is related to the pathological grade, metastasis and prognosis of renal cell carcinoma.
