目的探讨环保固定液聚羟基丙烯酸在Wade-Fite苯酚碱性品红法检测结核杆菌的应用价值。方法手术切除且术后病理证实为结核病标本96例,每例标本同一病变部位取材2块,用抽签法随机分为A、B组。A组采用4%中性缓冲甲醛固定,B组采用聚羟基丙烯酸固定,均制作切片96张。采用Wade-Fite苯酚碱性品红法检测结核杆菌,使用SPSS20.0分析比较A、B组切片结核杆菌阳性率的差异。结果 (1)生物显微镜下,两组切片染色结果:背景洁净、细胞轮廓清晰、胞核呈蓝色,背景着色为淡蓝色、结核杆菌呈鲜红色、对比度好。且同一病变部位B组切片显示结核杆菌数量显著多于A组。(2)A、B两组切片结核杆菌阳性率分别为58.3%和66.7%,两组间阳性率差异显著(χ2=6.125,P<0.05)。结论环保固定液聚羟基丙烯酸有潜能替代传统试剂4%中性缓冲甲醛应用于Wade-Fite苯酚碱性品红法检测结核杆菌。
Objective To investigate the application value of environmental protection fixative poly (hydroxyethyl) acrylic acid in the detection of Mycobacterium tuberculosis by Wade-Fite Phenol Alkaline Fuchsin method. Methods Surgical resection and histopathology were confirmed as 96 cases of tuberculosis specimens. Two specimens of the same lesion were taken from each specimen. The specimens were randomly divided into groups A and B by random sampling. A group of 4% neutral buffered formaldehyde fixation, B group using polyhydroxy acrylic acid fixed, were made 96 slices. Mycobacterium tuberculosis was detected by Wade-Fite Phenol-Alkaline Fuchsin method. The differences of the positive rate of Mycobacterium tuberculosis between group A and group B were analyzed by SPSS20.0. Results (1) Under biomicroscopy, two groups of sections were stained with clear background, clear cell outline, blue nuclei, pale blue background and bright red color with good contrast. And the same lesion B group showed a significant number of M. tuberculosis than A group. (2) The positive rates of Mycobacterium tuberculosis in groups A and B were 58.3% and 66.7% respectively, there was significant difference between the two groups (χ2 = 6.125, P <0.05). CONCLUSION Environmental protection liquid polyhydroxy acrylic acid has the potential to replace traditional reagent 4% neutral buffered formaldehyde for the detection of Mycobacterium tuberculosis by the Wade-Fite Phenol Alkaline Fuchsin method.
