目的探讨清肾颗粒对肾纤维化大鼠免疫系统及炎症反应的影响。方法采用单侧输尿管梗阻(UUO)建立肾纤维化模型,将38只SD大鼠随机分为清肾颗粒组(n=8)、百令胶囊组(n=8)、模型组(n=7)、假手术组(n=7)和正常组(n=8)。清肾颗粒组按0.3g/(100g·d)灌胃,百令胶囊组按0.06g/(100g·d)灌胃,模型组、假手术组及正常组每日给予等量温水灌胃,疗程3周。检测各组大鼠血尿素氮(BUN)、肌酐(Scr)和24h尿蛋白定量(U-Pro/24h)水平,采用流式细胞仪检测外周血CD_4~+T细胞、CD_8~+T细胞和Th17细胞含量,免疫组织化学法检测肾脏组织白介素-17(IL-17)、核因子-κB(NF-κB)p65和Ⅲ型胶原蛋白(Col-Ⅲ)相对含量。结果清肾颗粒组、百令胶囊组、模型组BUN、Scr、U-Pro/24h、外周血Th17细胞比例、CD_4~+/CD_8~+水平及肾组织中IL-17、NF-κBp65、Col-Ⅲ半定量评分均高于正常组和假手术组(P<0.05);治疗后与模型组比较,清肾颗粒组和百令胶囊组上述水平显著降低(P<0.01),清肾颗粒组优于百令胶囊组(P<0.05)。结论清肾颗粒可以通过调节UUO模型大鼠免疫系统功能,抑制炎症反应发挥抗肾纤维化作用。
Objective To investigate the effects of Qing Shen Granules on the immune system and inflammatory response in rats with renal fibrosis. Methods The model of renal fibrosis was established by unilateral ureteral obstruction (UUO). Thirty - eight SD rats were randomly divided into three groups: Qingxian Granule group (n = 8), Bailing capsule group (n = 8) ), Sham operation group (n = 7) and normal group (n = 8). The rats in the model group, the sham operation group and the normal group were given the same amount of warm water by intragastric administration with 0.3g / (100g · d), and the Bailing capsule group was fed with 0.06g / (100g · d) Treatment for 3 weeks. The levels of blood urea nitrogen (BUN), creatinine (Scr) and urinary protein (24 h) in each group were measured. The levels of CD_4 ~ + T cells and CD_8 ~ + T cells in peripheral blood were detected by flow cytometry Th17 cells and the relative content of IL-17, NF-κB p65 and Col-Ⅲ in kidneys were detected by immunohistochemical method. Results The levels of Th17 cells, CD_4 ~ + / CD_8 ~ + and the levels of IL-17, NF-κBp65, Col of renal tissue in BX, BRL, BUN, Scr and U-Pro / (P <0.05). Compared with the model group, the levels of Qing-Shen Granule and Bailing Capsule group were significantly lower than those of the model group (P <0.01) Better than Bailing capsule group (P <0.05). Conclusion Qing Shen Granules can regulate the immune system function of UUO model rats and inhibit the inflammatory reaction to exert anti-renal fibrosis.
