目的:研究RhoA、ROCK2在宫颈上皮内瘤样病变(cervical intraepithelial neoplasia,CIN)及宫颈癌组织中的表达,探讨其与CIN疾病及宫颈癌进展的相关性。方法:应用免疫组化S-P法,检测10例宫颈炎、30例不同级别CIN、30例宫颈癌组织中RhoA、ROCK2蛋白的表达情况,分析比较其表达与临床病理特征的关系。结果:①RhoA、ROCK2蛋白在宫颈炎组织中的表达率均为0%,在CIN宫颈组织中的表达率分别为36.7%、30.0%,在宫颈癌组织中的表达率分别为83.3%、76.7%,两者在宫颈癌组织中的表达水平高于CIN及宫颈炎组织(P<0.01),从CIN进展为宫颈癌,二者的阳性表达率呈递增趋势。②RhoA、ROCK2蛋白表达与宫颈癌病理组织分级及淋巴结转移相关(P<0.05),与宫颈癌患者的年龄、临床分期无明显相关关系(P>0.05)。③RhoA、ROCK2蛋白二者表达水平呈正相关关系(P=0.005)。结论:RhoA、ROCK2蛋白的表达水平与CIN疾病及宫颈癌进展呈正相关,检测RhoA、ROCK对了解CIN及宫颈癌生物学行为和评估预后具有一定的临床价值。
Objective: To investigate the expression of RhoA and ROCK2 in cervical intraepithelial neoplasia (CIN) and cervical cancer, and to investigate the relationship between RhoA and ROCK2 expression and the progression of cervical cancer. Methods: The expressions of RhoA and ROCK2 in 10 cases of cervicitis, 30 cases of different grades of CIN and 30 cases of cervical cancer were detected by immunohistochemical S-P method. The relationship between the expression of RhoA and ROCK2 protein was analyzed and compared with the clinicopathological features. Results: ①The expression rates of RhoA and ROCK2 in cervicitis were 0%, 36.7% and 30.0% in cervical intraepithelial neoplasia respectively, and 83.3% and 76.7% in cervical carcinoma respectively (P <0.01). The positive expression rates of CIN and cervicitis from cervical intraepithelial neoplasia (CIN) to cervical cancer showed an increasing trend. The expression of RhoA and ROCK2 protein was correlated with the pathological grade and lymph node metastasis of cervical cancer (P <0.05), but not with the age and clinical stage of cervical cancer (P> 0.05). There was a positive correlation between RhoA and ROCK2 protein expression (P = 0.005). Conclusion: The expression of RhoA and ROCK2 protein is positively correlated with the progression of CIN and cervical cancer. The detection of RhoA and ROCK has some clinical value in understanding the biological behavior of CIN and cervical cancer and evaluating the prognosis.
