目的观察尤瑞克林对脑缺血再灌注大鼠血管内皮生长因子(VEGF)和碱性成纤维细胞生长因子(b FGF)表达的影响。方法 SD大鼠随机分为假手术组、模型组和实验组。线栓法制作大脑中动脉脑缺血再灌注模型,实验组于再灌注后5 min静脉注射尤瑞克林3.5×10-3PNAU·kg-1。缺血2 h后再灌注48 h,断头取脑,免疫组织化学法和PCR法检测VEGF和b FGF的表达。结果与模型组相比,实验组大鼠脑组织中VEGF和b FGF的表达水平明显增高,差异均有统计学意义(P<0.05)。结论尤瑞克林对大鼠脑缺血再灌注损伤的保护作用可能与促进VEGF和b FGF的表达有关。
Objective To observe the effect of uracil on the expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (b FGF) in rats with cerebral ischemia-reperfusion. Methods SD rats were randomly divided into sham operation group, model group and experimental group. The model of middle cerebral artery occlusion (MCAO) was established by thread occlusion. The experimental group received intravenous injection of uracil 3.5 × 10-3 PNAU · kg-1 5 min after reperfusion. At 2 h after ischemia, the cells were reperfused for 48 h. The brain was decapitated. The expressions of VEGF and b FGF were detected by immunohistochemistry and PCR. Results Compared with the model group, the expression levels of VEGF and b FGF in the experimental group were significantly increased, the difference was statistically significant (P <0.05). Conclusion The protective effect of uracil against cerebral ischemia-reperfusion injury in rats may be related to the promotion of VEGF and b FGF expression.
