目的:观察坤宁安对围绝经期抑郁模型大鼠海马5HTR1A、5HTR2A受体亚型mRNA表达的影响。方法:采用孤养结合慢性轻度不可预见性应激造模法,观测围绝经期抑郁大鼠模型海马5HTR1A、5HTR2A受体亚型mRNA表达的变化。结果:抑郁模型组大鼠海马的5-HTR1A受体亚型mRNA表达降低(P<0.05),5-HTR2A受体亚型mRNA表达水平增高(P<0.05);坤宁安可使大鼠5-HTR1A受体亚型mRNA表达上调,使大鼠5-HTR2A受体亚型mRNA表达下调。结论:坤宁安影响抑郁模型大鼠海马5HTR1A、5HTR2A受体亚型mRNA的表达,可能是其治疗围绝经期抑郁症的机理之一。
Objective: To observe the effect of Kunningan on mRNA expression of 5HTR1A and 5HTR2A receptor subtypes in hippocampus of perimenopausal depression model rats. Methods: The changes of mRNA expression of 5HTR1A and 5HTR2A receptor subtypes in hippocampus of perimenopausal depression model were observed by solitary combined with chronic mild unpredictable stress modeling. Results: The mRNA expression of 5-HTR1A receptor subtype and the mRNA expression of 5-HTR2A receptor subtype in hippocampus of depression model group were significantly decreased (P <0.05) -HTR1A receptor subtypes mRNA upregulation, rat 5-HTR2A receptor subtype mRNA expression down regulation. Conclusion: Kunningan can affect the expression of 5HTR1A, 5HTR2A receptor subtypes mRNA in the hippocampus of the depression model rats, which may be one of its mechanisms in the treatment of perimenopausal depression.
