目的:检测一氧化氮合酶(NOS)抑制剂非对称性二甲基精氨酸(ADMA)对原发性高血压(EH)患者外周血单核细胞趋化活性的影响。方法:收集42例EH患者(EH组)和40例门诊健康体检者(对照组)。检测血浆中ADMA和NO水平、血管假性血友病因子(vWF)、白细胞介素8(IL-8)和肿瘤坏死因子(TNF)-α浓度,并观察ADMA对外周血单核细胞IL-8受体CXCR2 mRNA表达的影响。结果:与对照组比较,EH患者血浆中AD-MA和vWF水平升高、NO生成减少(P<0.05),同时血中IL-8和TNF-α水平显著升高(P<0.05)。EH患者分离的外周血单核细胞CXCR2 mRNA表达明显高于对照组,外源性的ADMA 30μmol/L孵育4 h后可进一步上调CXCR2 mRNA的表达(P<0.05)。结论:EH患者血中升高的ADMA可能增加外周血单核细胞的趋化活性,促进动脉粥样硬化的发生。
Objective: To investigate the effect of asymmetric dimethylarginine (ADMA), a nitric oxide synthase (NOS) inhibitor, on the chemotactic activity of peripheral blood monocytes in patients with essential hypertension (EH). Methods: 42 patients with EH (EH group) and 40 healthy subjects (control group) were collected. The levels of ADMA and NO, vWF, IL-8 and TNF-α in plasma were measured. The effects of ADMA on the levels of IL- 8 receptor CXCR2 mRNA expression. Results: Compared with the control group, the levels of AD-MA and vWF in plasma of patients with EH increased and the production of NO decreased (P <0.05). The levels of IL-8 and TNF-α in blood increased significantly (P <0.05). The CXCR2 mRNA expression of peripheral blood mononuclear cells isolated from patients with EH was significantly higher than that of the control group, and the expression of CXCR2 mRNA was up-regulated after treated with exogenous ADMA 30 μmol / L for 4 h (P <0.05). CONCLUSIONS: Elevated ADMA in EH patients may increase the chemotactic activity of peripheral blood mononuclear cells and promote the development of atherosclerosis.
