目的探讨体外模拟二氧化碳气腹对宫颈恶性肿瘤细胞生长转移能力的影响。方法体外培养人宫颈癌Hela细胞株,以CO2分别处理1,2,3h后,置于常规培养箱中分别培养24,48,72h,采用实时荧光定量PCR法及免疫细胞化学法检测各组Hela细胞VEGF-C、MMP9 mRNA和蛋白的表达。结果 CO2处理后Hela细胞VEGF-C、MMP9 mRNA和蛋白的表达均显著高于对照组(P<0.01);在不同CO2处理时间及培养时间下,VEGF-C、MMP9 mRNA和蛋白的表达均有差异(P<0.01)。结论 CO2环境可促进Hela细胞生长转移因子VEGF-C、MMP9表达增加,从而增强Hela细胞的生长转移能力。
Objective To investigate the effects of carbon dioxide pneumoperitoneum on the growth and metastasis of cervical malignant cells in vitro. Methods Hela cell lines were cultured in vitro and treated with CO2 for 1, 2 and 3 hours respectively. The cells were cultured in a routine incubator for 24, 48 and 72 hours respectively. Real-time fluorescent quantitative PCR and immunocytochemistry were used to detect Hela Cell VEGF-C, MMP9 mRNA and protein expression. Results The mRNA and protein expressions of VEGF-C and MMP9 in Hela cells were significantly higher than those in control group (P <0.01). The expressions of VEGF-C and MMP9 mRNA and protein in different CO2 treatment time and culture time Difference (P <0.01). Conclusion CO2 environment can promote the expression of VEGF-C and MMP9 in Hela cells and enhance the growth and metastasis of Hela cells.
