通过发现内皮素对肺动脉及肺动脉平滑肌细胞功能的可能影响 ,以便进一步探讨内皮素在缺氧性肺动脉高压发病中的作用。取大鼠肺动脉制成 3mm长的血管环 ,让血管环在最佳静息张力下平衡一段时间后 ,按累积法向浴槽内加入内皮素 1,用二道生理记录仪记录不同浓度的内皮素 1对血管环收缩功能影响。另采取单层贴壁培养家兔肺动脉中膜平滑肌细胞 ,在低血清培养条件下 ,加入不同浓度的内皮素 1,分别用氚标胸腺嘧啶和氚标亮氨酸掺入率来评估内皮素 1对肺动脉平滑肌细胞DNA和蛋白质合成的促进作用。结果发现 :①内皮素 1能引起肺动脉剂量依赖性收缩 (r=0 .935 ,P <0 .0 5 ) ,在 ( 1~ 12 )× 10 - 9mol L浓度范围内 ,内皮素 1所引起的血管张力迅速上升 ,由 40± 6mg上升至 347± 12mg,当浓度高于 12× 10 - 9mol L时 ,收缩张力上升速度减慢。②当内皮素 1浓度为 1× 10 - 1 1 ~ 1× 10 - 8mol L时 ,肺动脉平滑肌细胞氚标胸腺嘧啶脱氧核苷和氚标亮氨酸掺入率呈现剂量依赖性增加 (r分别为 0 .75 6和 0 .840 ,P <0 .0 5 ) ,其EC5 0 分别为 10 .87mol L和 8.5 4mol L。结果提示 ,内皮素具有较强的肺血管活性
By discovering the possible effect of endothelin on pulmonary artery and pulmonary artery smooth muscle cell function in order to further explore the role of endothelin in the pathogenesis of hypoxic pulmonary hypertension.
