胃粘膜细胞尤其是壁细胞已经发现有C-erb B3基因产物中等量表达。对胃癌细胞株用Northernblot分析发现有6.2-kb的C-erb B3转录基因表达,在MKN45胃癌细胞系有1.4kb转录基因和6.2-kb转录基因表达,并且这2种转录基因的表达水平是相同的。通过末端cDNA快速扩增法将1.4kb C-erb B3转录基因对应的erb B3-S核酸片断得到克隆,并对erb B3-S cDNA进行序列分析,发现1.4-kb C-erb B3 cRNA编码一种分泌受体。对c-erbB3部分基因结构分析发现这种分泌受体特异性的外显与C-erb B3基因5′端的内含子完全相同,表明C-erb B3基因编码一种具有酪氨酸激酶活性的膜受体。
Gastric mucosal cells, especially parietal cells, have been found to have an equivalent amount of C-erb B3 gene product. Northern blot analysis of the gastric cancer cell line revealed a 6.2-kb C-erb B3 transcriptional gene expression in the MKN45 gastric cancer cell line with a 1.4 kb transcriptional gene and a 6.2-kb transcriptional gene expression, and the expression levels of the two transcriptional genes were the same of. Cloning of the erb B3-S nucleic acid fragment corresponding to the 1.4 kb C-erb B3 transcript gene by rapid amplification of the terminal cDNA and sequencing of the erb B3-S cDNA revealed that the 1.4-kb C-erb B3 cRNA encodes a Secretion of receptors. Structural analysis of part of the c-erbB3 gene revealed that the specificity of this secretion receptor was exactly the same as that of the 5 ’end of the C-erb B3 gene, indicating that the C-erb B3 gene encodes a tyrosine kinase- Membrane receptors.
