目的:初步探索葡萄柚黄酮(PTFCs)联合三氧化二砷(As2O3)协同诱导白血病Kasumi-1细胞株凋亡及凋亡相关机制。创新点:采用生物酶法与超声波辅助提取技术结合的方法从天然植物葡萄柚中提取和纯化出黄酮组分已取得国家发明型专利,发现其具有明显的抑制白血病细胞增殖作用,并且与As2O3存在协同作用。方法:通过MTT法验证其抑制Kasumi-1细胞增殖作用,通过流式细胞术、Hoechst 33258染色、蛋白质印迹(Western blot)等技术探索其相关作用机制。结论:发现PTFCs可以协同As2O3诱导Kasumi-1细胞凋亡。
Objective: To explore the mechanisms of apoptosis and apoptosis associated with synergistic induction of leukemia cell line Kasumi-1 by grapefruit flavonoids (PTFCs) and arsenic trioxide (As2O3). Innovative point: the use of bio-enzyme method and ultrasonic-assisted extraction method from the natural plant grapefruit extract and purification of flavonoids has made the national invention patents found that it has a significant inhibitory effect on leukemia cells, and with the presence of As2O3 Synergy. Methods: The proliferation of Kasumi-1 cells was verified by MTT assay. The related mechanism was explored by flow cytometry, Hoechst 33258 staining and Western blot. Conclusion: It was found that PTFCs can induce Kasumi-1 cell apoptosis in coordination with As2O3.
