目的探讨少突胶质细胞系基因(oligodendrocyte lineage gene)Olig2和同源盒基因(Ho-meobox)Hb9在小鼠胚胎脊髓发育过程中的表达情况。方法选择小鼠的9.5~13.5d胚胎脊髓,并分为E9.5d组、E10.5d组、E12.5d组、E13.5d组;采用免疫荧光方法检测Olig2和Hb9在胎鼠脊髓中的分布,同时用流式细胞仪检测分别标记Olig2和Hb9的阳性细胞百分率。结果免疫荧光染色显示在这5组胎鼠脊髓的腹侧都有Olig2和Hb9的表达,流式细胞仪检测显示E9.5d组Olig2阳性细胞百分率高于其它4组,差异有统计学意义(P<0.05),E11.5d组Hb9的阳性细胞百分率高于其它4组,差异有统计学意义(P<0.05)。结论Olig2和Hb9可能与胎鼠脊髓运动前体细胞和运动神经元的发育有关。
Objective To investigate the expression of oligodendrocyte lineage gene Olig2 and homeobox gene Hb9 in mouse embryonic spinal cord. Methods The embryonic spinal cord of 9.5 ~ 13.5 days was selected and divided into E9.5d group, E10.5d group, E12.5d group and E13.5d group. The distribution of Olig2 and Hb9 in fetal rat spinal cord was detected by immunofluorescence Meanwhile, the percentage of positive cells labeled with Olig2 and Hb9 were detected by flow cytometry. Results Immunofluorescence staining showed that Olig2 and Hb9 were expressed on the ventral side of fetal rat spinal cord in these 5 groups. Flow cytometry showed that the percentage of Olig2 positive cells in E9.5d group was higher than that in the other 4 groups (P <0.05). The percentage of Hb9 positive cells in E11.5d group was higher than that in other 4 groups (P <0.05). Conclusion Olig2 and Hb9 may be related to the development of somatic cells and motor neurons in the fetal rat spinal cord.
