目的探究健肝颗粒抗肝纤维化的作用机制。方法选取100只雄性SD大鼠,随机分组后以四氯化碳腹腔内注射建立大鼠肝纤维化模型,造模后施以由低、中、高剂量健肝颗粒,以秋水仙碱为阳性对照。行苏木素-伊红、Masson染色,免疫组织化学染色检测肝组织转化生长因子β1(TGFβ1)、RT-PCR法检测肝组织中核因子-kB(NF-kB)mRNA的表达。结果健肝颗粒能降低大鼠肝纤维化病理评分,改善肝纤维化病理损伤;降低炎症因子TGFβ1、NF-kB mRNA在肝组织中的表达,尤其以低剂量健肝颗粒组效果明显(P<0.01)。结论健肝颗粒组能有效改善大鼠肝纤维化的病理改变;减少TGFβ1、NF-kB mRNA表达可能是其相关作用机制。
Objective To investigate the mechanism of Jiangan Granule in preventing liver fibrosis. Methods One hundred male Sprague-Dawley rats were randomly divided into three groups: rat model of hepatic fibrosis induced by intraperitoneal injection of carbon tetrachloride (CCl4) Control. Transforming growth factor β1 (TGFβ1) was detected by hematoxylin-eosin, Masson staining and immunohistochemical staining. The expression of nuclear factor-kB (NF-kB) mRNA was detected by RT-PCR. Results Jiangan Granules could reduce the pathological score of liver fibrosis, improve the pathological damage of hepatic fibrosis, and reduce the expression of inflammatory factors TGFβ1 and NF-kB mRNA in liver tissue, especially in low dose Jiangan granule group (P < 0.01). Conclusion Jiangan granule group can effectively improve the pathological changes of liver fibrosis in rats; reduce TGFβ1, NF-kB mRNA expression may be related mechanism.
