实验以花魔芋种子为材料,研究无菌播种诱导萌发试管苗技术,以获得无菌试管植株,并由此获得外植体,从而解决魔芋组织培养过程中污染率及褐变率高的问题。实验结果表明,播种于MS培养基中的魔芋种子经培养30 d即可获无菌幼苗,将无菌幼苗的叶柄、叶片接种于MS+1.0 mg/L 6-BA+1.5 mg/L NAA培养基中培养,叶柄、叶片愈伤组织诱导率可达64%、58%,幼苗分化率可达74%、62%。无根芽苗接种于1/2 MS+0.5 mg/L NAA培养基中进行生根培养,14 d可长出根系,其生根率达97%。
In the experiment, the seeds of konjac were used to study the technology of germ-free in vitro seed germination in order to obtain sterile test tube plants, and to obtain explants, thus solving the problem of high pollution rate and high browning rate in konjak tissue culture. The results showed that seedlings of Amorphophallus konjac were sown in MS medium for 30 days and petiole and leaves of sterile seedlings were inoculated into MSA 1.0 mg / L 6-BA + 1.5 mg / L NAA In the basal medium, the induction rates of petiole and leaf callus could reach 64% and 58%, respectively. The rate of seedling differentiation reached 74% and 62% respectively. Rooted shoots were inoculated on 1/2 MS + 0.5 mg / L NAA medium for rooting, and on the 14th day, roots could be grown with rooting rate of 97%.
