采用改良的高效液相色谱方法,以贡菊为对照,测定了不同品系昆仑雪菊中绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸及槲皮苷的含量。结果表明,较适宜的高效液相色谱条件为Alitima HP C18 5μm色谱柱(250mm×4.6 mm),以乙腈-0.1%磷酸水溶液梯度洗脱,流速1 m L·min-1,检测波长为350 nm,柱温40℃,进样量5μL。不同品系的昆仑雪菊与贡菊主要活性成分相似,但其含量差异显著。用该方法测得昆仑雪菊和贡菊中的绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸及槲皮苷的含量均高于药典规定的含量标准,其中昆仑雪菊中绿原酸、木犀草苷和槲皮苷的含量显著高于供试贡菊。
The contents of chlorogenic acid, luteolin, 3,5-O-dicaffeoylquinic acid and quercitrin in different strains of Kunluoshuju were determined by a modified high performance liquid chromatography with Gongju as control. The results showed that the most suitable HPLC conditions were Alitima HP C18 5 μm column (250 mm × 4.6 mm), gradient elution with acetonitrile-0.1% phosphoric acid solution, flow rate 1 m L · min-1 and detection wavelength of 350 nm , Column temperature 40 ℃, injection volume 5μL. Kunlun snowdrops of different strains were similar to the main active ingredients of Gongju, but their contents were significantly different. Using this method, the contents of chlorogenic acid, luteolin, 3,5-O-dicaffeoylquinic acid and quercitrin in Kunlun Xueju and Gongju were all higher than those determined by Pharmacopoeia, of which Kunlun Snow chrysanthemum chlorogenic acid, luteolin and quercitrin content was significantly higher than that for tribute chrysanthemum.
