目的:筛选卵巢癌铂类耐药细胞和敏感细胞间的差异蛋白质。方法:提取卵巢癌耐卡铂细胞SKOV3/CB和亲本细胞SKOV3的总蛋白,用二维液相色谱技术(ProtemeLabTM PF-2D)分离、筛选差异蛋白,用电喷雾离子化串联质谱(ESI-MS/MS)分析鉴定蛋白质。结果:共分离筛选出差异蛋白54个,其中SK-OV3/CB表达上调的34个,SKOV3表达上调的20个。初步鉴定出5个SKOV3/CB表达上调的差异蛋白,即硫氧还原蛋白(thioredoxin,Trx)、Myosin regulatorylight polypeptide 9(MYL9)、AX887247NID、stanniocalcin homolog、human regu-lator of G-protein signaling 5(RGS-5)。这些蛋白涉及氧化还原、凋亡调控、甲基化及信号传导等多方面。结论:卵巢癌铂类耐药细胞和敏感细胞间存在蛋白表达差异,这些差异蛋白可能通过多种机制介导了卵巢癌对铂类的耐药。
OBJECTIVE: To screen differential proteins between platinum-resistant and sensitive cells in ovarian cancer. Methods: Total proteins of ovarian cancer SKOV3 / CB cells and SKOV3 cells were extracted and separated by two-dimensional liquid chromatography (ProtemeLabTM PF-2D). The differential proteins were screened by electrospray ionization tandem mass spectrometry (ESI-MS / MS) analysis to identify proteins. RESULTS: A total of 54 differentially expressed proteins were screened out, of which 34 were upregulated in SK-OV3 / CB and 20 were up-regulated in SKOV3. Five proteins that were upregulated in SKOV3 / CB were initially identified, namely thioredoxin (Trx), Myosin regulatory light polypeptide 9 (MYL9), AX887247NID, stanniocalcin homolog, human regu-lator of G-protein signaling 5 -5). These proteins are involved in many aspects such as redox, apoptosis regulation, methylation and signal transduction. CONCLUSION: There are differences in protein expression between platinum-resistant and sensitive cells in ovarian cancer. These differential proteins may mediate platinum resistance in ovarian cancer through a variety of mechanisms.
