目的:建立同时测定秋海棠、中华秋海棠的块茎、茎、叶中5种成分(原儿茶酸、原儿茶醛、儿茶素、表儿茶素和芦丁)含量的UPLC方法。方法:采用ACQUITY UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm);以甲醇-0.1%甲酸溶液为流动相,梯度洗脱;流速0.2 m L/min;检测波长280 nm;柱温30℃。结果:所有样品5个成分均达到基线分离,线性关系良好,原儿茶酸、原儿茶醛、儿茶素、表儿茶素、芦丁的加样回收率分别为:99.45%、96.92%、99.69%、98.42%、99.75%(RSD均小于5%)。结论:该方法简便、准确、灵敏度高,能较好的反映秋海棠、中华秋海棠的质量情况,为秋海棠、中华秋海棠的后续开发利用提供参考依据。
OBJECTIVE: To establish an UPLC method for the simultaneous determination of five components (protocatechuic acid, protocatechuic aldehyde, catechin, epicatechin and rutin) in tuber, stem and leaf of begonias and Chinese begonias. METHODS: ACQUITY UPLC HSS T3 column (100 mm × 2.1 mm, 1.8 μm) was used. The mobile phase consisted of methanol-0.1% formic acid as mobile phase with a gradient of 0.2 m L / min. The detection wavelength was set at 280 nm. ℃. Results: The five components of all samples reached baseline separation with good linearity. The recoveries of protocatechuic acid, protocatechuic aldehyde, catechin, epicatechin and rutin were 99.45% and 96.92%, respectively. , 99.69%, 98.42%, 99.75% (RSD less than 5%). Conclusion: The method is simple, accurate and sensitive. It can better reflect the quality of Begonia begonia and Chinese begonia, and provide a reference for the follow-up development and utilization of begonias and Chinese begonias.
