研究了大鼠卵巢黄体中胰岛素样生长因子-Ⅰ受体(IGF-ⅠR)与促黄体激素受 体(LHR)的表达关系及调节.在动情周期中, LHR阳性黄体能表达高水平的 IGF- ⅠR mRNA.动情期黄体的 LHR表达水平降低,而 IGF-ⅠR表达仍较高.假孕第 1天黄体中 LHR表达较低,第8天达最高水平,第14天表达已降低到检测不到的程度.与LHR表 达不同,在第 1天假孕黄体中 IGF-ⅠR的表达已较高,此后至第 14天在黄体中均检测 到 IGF- ⅠR信号孕鼠黄体同时表达 LHR和 IGF- ⅠR mRNA. IGF- Ⅰ刺激 LHR mRNA 在黄体中的表达.前列腺素(PG)F2a抑制 IGF- ⅠR和LHR的表达.PGE2能消除PGF2a对 IGF-IR和 LHR mRNA表达的抑制作用.上述结果提示:IGF-I可能通过其受体的表达变化参与调节黄体功能、维持黄体结构.PGF2a抑制 IGF- IR的表达可能是黄体萎缩 的机制之一.
The relationship and regulation of insulin-like growth factor-I receptor (IGF-IR) and luteinizing hormone receptor (LHR) expression in the rat luteal ovary were studied. During the estrous cycle, LHR-positive luteal can express high levels of IGF-IR mRNA. Luteal LHR LHR expression decreased, while IGF-IR expression is still high. The LHR expression in the corpus luteum was lower on the first day of pseudopregnancy and reached the highest level on the 8th day. The expression on the 14th day had been reduced to the undetectable level. Different from LHR expression, the expression of IGF-IR was higher on day 1 of pseudopregnant corpus luteum and LHR and IGF-IR mRNA were detected in the corpus luteum of all pregnant rats. The expression of LHR mRNA stimulated by IGF-Ⅰ in corpora lutea. Prostaglandin (PG) F2a inhibits the expression of IGF-IR and LHR. PGE2 abolished the inhibitory effect of PGF2a on IGF-IR and LHR mRNA expression. These results suggest that: IGF-I may be involved in the regulation of the function of the corpus luteum through its receptor expression changes, maintain corpus luteum structure. PGF2a inhibition of IGF-IR expression may be one of the mechanisms of luteal atrophy.
