[目的]探讨小儿哮喘与血浆环核苷酸的关系及黎氏哮喘Ⅰ号方(由麻黄、桂枝、五味子、鹅管石、当归、毛冬青、五指毛桃根等组成)治疗哮喘的作用机理。[方法]将70例哮喘患儿随机分为黎氏哮喘Ⅰ号方治疗组(A)47例和氨茶碱对照组(B)23例,并设10例健康儿童对照组(C),应用放射免疫法观察各组治疗前后血浆cAMP(环磷酸腺苷)、cGMP(环磷酸鸟苷)水平,并在哮喘豚鼠模型上进行验证。同时利用透射式电子显微镜观察黎氏哮喘Ⅰ号方对哮喘豚鼠肺超微结构变化的影响。[结果]治疗后A、B两组哮喘儿童cAMP、cGMP水平均有改善,cAMP/cGMP比值上升(与治疗前相比,均P<0.01)。动物实验表明,治疗后中药高剂量组cAMP/cGMP比值改善优于氨茶碱组(P<0.01)。黎氏哮喘Ⅰ号方连续治疗7 d后,哮喘豚鼠肺毛细血管壁增厚程度减轻,无胶原纤维增生。[结论]黎氏哮喘Ⅰ号方能提高哮喘患儿及哮喘豚鼠cAMP/cGMP比值,减轻哮喘豚鼠肺组织的炎症程度。
[Objective] To investigate the relationship between pediatric asthma and plasma cyclic nucleotides and the mechanism of action of Asthma Asthma Ⅰ (composed of ephedra, cassia twig, Schisandra chinensis, gooseneck, Angelica sinensis, Mao dongqing, five fingers peach root etc.) . [Methods] Seventy patients with asthma were randomly divided into three groups: 47 cases of AML (A group) and 23 cases of aminophylline control group (B), and 10 healthy children control group (C) Radioimmunoassay was used to observe the levels of plasma cAMP (cyclic AMP) and cGMP (cyclic guanosine monophosphate) before and after treatment in each group and validated on the model of asthmatic guinea pigs. Meanwhile, transmission electron microscopy was used to observe the effects of Li's Asthma Part I on lung ultrastructure in asthmatic guinea pigs. [Results] After treatment, the levels of cAMP and cGMP in both asthmatic children and the cAMP / cGMP increased in both A and B groups (all P <0.01 compared with those before treatment). Animal experiments show that, after treatment, high-dose group of Chinese medicine cAMP / cGMP ratio improved better than aminophylline group (P <0.01). After treatment of Li Asthma Ⅰ for 7 days, the thickening of pulmonary capillary wall in asthmatic guinea pigs was alleviated and no collagen fibers proliferated. [Conclusion] Lebanon Asthma Ⅰ can improve the cAMP / cGMP ratio in asthmatic children and asthmatic guinea pigs, and reduce the degree of inflammation in the lung tissue of asthmatic guinea pigs.
