目的分析丙型肝炎病毒(HCV)非结构基因5b(NS5b)区部分核苷酸序列特征。方法以逆转录巢式聚合酶链反应获得位于7918位至8293位间的cDNA片段,克隆后测定重组子序列,计算机分析其特征。结果获得了376碱基对长的HCVNS5b基因(HC-P39),与HCV-1,HC-C2,HCV-J,HCV-BK,HCV-J6及HC-J8相比,核苷酸水平的同源性分别为76.97%,88.92%,87.75%,89.21%,63.85%及64.43%。在核苷酸8111位至核苷酸8124位发现一个新的移码突变,并因此在核苷酸8122位形成一个终止突变。结论在HCVNS5b区发现一个新的移码突变。
Objective To analyze the partial nucleotide sequence of non-structural gene 5b (NS5b) of hepatitis C virus (HCV). Methods cDNA fragments from 7918 to 8293 were obtained by reverse transcriptase nested polymerase chain reaction (PCR). The cloned recombinant DNA fragments were sequenced and their characteristics were analyzed by computer. As a result, a 376 base pair long HCV NS5b gene (HC-P39) was obtained. Compared with HCV-1, HC-C2, HCV-J, HCV-BK, HCV-J6 and HC-J8, Source was 76.97%, 88.92%, 87.75%, 89.21%, 63.85% and 64.43% respectively. A new frameshift mutation was found at nucleotide 8111 to nucleotide 8124 and therefore a stop mutation at nucleotide 8122. Conclusion A new frameshift mutation was found in HCV NS5b region.
