目的探讨白花丹醌对人黑色素瘤A375细胞增殖和凋亡的影响及其可能机制。方法用不同浓度白花丹醌处理A375细胞后,采用MTT法和流式细胞术分别检测白花丹醌对A375细胞增殖和凋亡的影响,Western blot检测JAK-2、信号传导与转录激活因子3(STAT-3)、B细胞淋巴瘤-白血病2(Bcl-2)的表达。结果随着白花丹醌浓度升高,A375细胞增殖抑制率和凋亡率明显增加,JAK-2、STAT-3磷酸化水平以及Bcl-2蛋白表达均显著降低(P<0.05)。结论白花丹醌能抑制A375细胞增殖并促进凋亡,可能与其抑制了JAK-2/STAT-3信号通路蛋白的活化有关。
Objective To investigate the effect of plumbagin on proliferation and apoptosis of human melanoma A375 cells and its possible mechanism. Methods A375 cells were treated with different concentrations of plumbagin. MTT assay and flow cytometry were used to detect the effects of plumbagin on the proliferation and apoptosis of A375 cells. The expressions of JAK-2, signal transducers and activators of transcription 3 STAT-3), B-cell lymphoma-leukemia 2 (Bcl-2). Results As the concentration of plumbagin was increased, the proliferation inhibition rate and apoptosis rate of A375 cells were significantly increased. The phosphorylation of JAK-2, STAT-3 and the expression of Bcl-2 protein were significantly decreased (P <0.05). Conclusions Plumbagin can inhibit the proliferation and promote the apoptosis of A375 cells, which may be related to its inhibition of the activation of JAK-2 / STAT-3 signaling pathway.
