目的探讨六味地黄丸(TJ-87)对产后母性束缚应激致仔代生长迟缓的影响。方法应用ICR小鼠建立产后母性束缚应激模型,产后第2天将母鼠移离仔鼠,给予母鼠每天1次每次3小时的束缚应激,持续至产后3周断乳时结束。本研究以仔鼠为研究对象:①研究断乳时模型组及正常对照组,观察两组仔鼠的体质量、身长、股骨长、血中胰岛素样生长因子(IGF-1)水平及脑垂体生长激素GH mRNA表达情况;②研究断乳后给予六味地黄丸(TJ-87)大、中、小剂量各治疗组(药物与饲料混合浓度为1.8%,1.2%,0.6%剂量组,连续给药8周)与模型组,观察治疗后各组仔鼠的体质量、身长、股骨长、血中IGF-1水平及脑垂体GH mRNA表达情况。结果①与正常对照组比较,产后3周断乳时模型组仔鼠表现出低体质量、低身长及低股骨长,同时模型组仔鼠血浆IGF-1水平降低,垂体GH mRNA相对表达低下,差异均有统计学意义(P<0.01)。②经六味地黄丸治疗8周后,与模型组比较,六味地黄丸大、中剂量组仔鼠身长及股骨长增加,同时六味地黄丸大、中剂量组仔鼠血浆IGF-1水平升高,其垂体GH mRNA相对表达也升高,差异有统计学意义(P<0.05,P<0.01);六味地黄丸大、中、小剂量组仔鼠体质量差异均无统计学意义(P>0.05)。结论六味地黄丸可能通过GH-IGF-1通路改善产后母性束缚应激仔代的生长。
Objective To investigate the effect of Liuwei Dihuang Pill (TJ-87) on postpartum maternal restraint stress induced growth retardation. Methods ICR mice were used to establish the model of postpartum maternal restraint stress. The maternal rats were removed from the pups on the second day postpartum, and the maternal rats were restrained for 3 hours once a day for 3 hours until the end of weaning. In this study, the offspring rats as the research object: ① weanling model group and normal control group, two groups of pups weight, length, femur length, blood insulin-like growth factor (IGF-1) levels and pituitary (2) To study the effect of Liuwei Dihuang Pill (TJ-87) on the expression of growth hormone GH mRNA; (2) Drug for 8 weeks) and model group. The body weight, body length, femur length, blood IGF-1 level and pituitary GH mRNA expression in each group were observed after treatment. Results ① Compared with the normal control group, the offspring of the model group showed low body weight, low body length and low femur length at 3 weeks postpartum, meanwhile the level of plasma IGF-1 in the model group was decreased, the relative expression of pituitary GH mRNA was low, The differences were statistically significant (P <0.01). After treatment with Liuweidihuang Pill for 8 weeks, compared with the model group, the body weight and femoral length increased in both large and medium doses of Liuweidihuangwan group, while the levels of IGF-1 in the large and medium doses of Liuweidihuangwan group were increased, The relative expression of GH mRNA in pituitary was also increased, the difference was statistically significant (P <0.05, P <0.01). There was no significant difference in body weight among the large, middle and small doses of Liuweidihuangwan group (P> 0.05) . Conclusion Liuweidihuangwan may improve the growth of postpartum maternal restraint stress generation through GH-IGF-1 pathway.
