目的: 了解rh T N Fα Db50 G 体内体外抗肿瘤活性。方法: 体外采用 M T T 法测rh T N Fα Db50 G 对 M K N45 细胞的 I C50 , 体内采用动物移植性肿瘤检测其抗肿瘤活性。结果: M T T法测定结果表明rh T N Fα Db50 G 对一定浓度的人胃癌细胞株 M K N45 有较强的抑制增殖作用, 其 I C50 为4 .461 ×10 - 2μg/ ml( 相关系数γ= 0 .9546) 。体内研究表明rh T N Fα Db50 G 对小鼠 S180 实体瘤和小鼠 Ehrlich 氏实体瘤有较强的抑制作用,其抑制率分别为29 .3 % (iv) 和40 .07 % (iv) 。rh T N Fα Db50 G 对小鼠黑色素瘤 B16 的肺转移有很强的抑制作用, 其抑制率为60 .41 % (iv) 。结论: 人重组肿瘤坏死因子rh T N Fα Db50 G 在体内体外均有一定的抗肿瘤作用。
Objective: To understand the anti-tumor activity of rh TN FαDb50 G in vitro and in vivo. METHODS: In vitro, MTT method was used to measure the I C50 of rhNNAFαDb50G against MKN45 cells. Animal transplanted tumors were used to examine their antitumor activity. RESULTS: The MTT assay results showed that rh-TN Fα-Db-50G had a strong inhibitory effect on the proliferation of MKN-45 human gastric cancer cell lines, and its I C50 was 4. 461 × 10 - 2μg/ml (correlation coefficient γ = 0 .9546). In vivo studies showed that rh-TN FαDb50 G had a strong inhibitory effect on S180 solid tumors in mice and Ehrlich solid tumors in mice. The inhibition rates were 29. 3% (iv) and 40. 07 % (iv). Rh T N Fα Db 50 G has a strong inhibitory effect on the lung metastasis of mouse melanoma B16 with an inhibition rate of 60. 41% (iv). Conclusion: Human recombinant tumor necrosis factor rh T N Fα Db 50 G has a certain anti-tumor effect in vitro and in vivo.
