观察铁超负荷对Wistar大鼠胰岛细胞功能的影响。雄性Wistar大鼠65只随机分为四组:A组(铁干预组)、B组(Na2NTA对照组)、C组(铁干预加去铁组)和D组(空白对照组)。于实验开始时、第5周、第10周时行腹腔注射葡萄糖耐量试验,检测血清铁、血清铁蛋白水平,取胰腺组织做电镜检查;检测胰岛中胰岛素、胰高糖素、生长抑素的水平,测定大鼠胰岛面积。实验第10周时发现,A、C组糖负荷前血糖较B、D组有增高趋势,2h血糖也较B、D组增高;电镜示A组大鼠β细胞胰岛素分泌颗粒明显减少,同时其胰岛的平均面积也明显小于其它组;各组间胰岛细胞的组成没有明显变化。铁超负荷使大鼠糖耐量受损,同时胰岛β细胞分泌胰岛素能力也有降低趋势,但给予去铁处理对大鼠的糖代谢具有一定的保护作用。
To observe the effect of iron overload on islet cell function in Wistar rats. Sixty-five male Wistar rats were randomly divided into four groups: A group (iron intervention group), B group (Na2NTA control group), C group (iron intervention plus iron group) and D group (blank control group). At the beginning of the experiment, glucose tolerance test was performed by intraperitoneal injection at the fifth week and the tenth week, serum iron and serum ferritin levels were measured, and the pancreatic tissue was examined by electron microscopy. The levels of insulin, glucagon and somatostatin in the islets were measured, Rat islet area was measured. At the 10th week of the experiment, the blood glucose of group A and C before glucose loading tended to be higher than that of group B and D, and the blood glucose of group 2h was also higher than that of group B and D at the 10th week. The electron microscopy showed that the fraction of insulin secreting from β cells was significantly decreased The average islet area was also significantly smaller than the other groups; the composition of islet cells did not change significantly among the groups. Iron overload impaired glucose tolerance in rats, while pancreatic β-cell insulin secretion also decreased, but to give iron treatment on rats with glucose metabolism has a protective effect.
