目的: 检测人工合成血管内皮细胞生长因子(VEGF)表位模拟 7肽特异抑制VEGF促进人脐静脉内皮细胞(HUVEC)生长的活性。方法: 通过噬菌体展示技术, 筛选获得了表达模拟肽的阳性噬菌体克隆, 经测序得到VEGF表位模拟肽序列: GWYYDAX。人工合成该短肽, 用体外细胞培养的方法检测其对HUVEC的生长抑制作用。结果: 表位模拟肽GWYYDAX可剂量依赖性抑制VEGF促进HUVEC生长的活性。结论: VEGF表位模拟肽可能模拟了VEGF的部分结构, 竞争结合HUVEC细胞VEGF受体 (KDR), 阻断了VEGF VEGFR的信号传递而抑制细胞生长, 具有潜在的应用前景。
OBJECTIVE: To detect the activity of artificial synthetic vascular endothelial growth factor (VEGF) epitope-mimicking 7-peptide to specifically inhibit the growth of human umbilical vein endothelial cells (HUVECs). Methods: The positive phage clones expressing the mimetic peptide were screened by the phage display technique. The mimic peptide sequence of the VEGF epitope was obtained by sequencing: GWYYDAX. The short peptide was synthesized by human in vitro and the growth inhibition of HUVEC was tested by in vitro cell culture. Results: Epitope mimic peptide GWYYDAX inhibited the growth of HUVECs in a dose-dependent manner. CONCLUSION: The mimic epitopes of VEGF epitopes may mimic the partial structure of VEGF and may compete with the VEGF receptor (KDR) of HUVEC cells to block the signal transduction of VEGF VEGFR and inhibit the cell growth. Therefore, it has potential application prospect.
