用碘化还原法将溴脱氧尿嘧啶(BudR)与牛r—球蛋白(BGG)偶联作为抗原(BudR—BGG)免疫BALB/c小鼠,取脾细胞与小鼠骨髓瘤细胞(653)进行融合,经放射免疫分析法(RIA)筛选和克隆化后,获得四株阳性杂交瘤Ⅱ_1B_7、Ⅱ_4B_4、Ⅰ_3F_(11)和Ⅱ_1H_7。杂交瘤细胞分别腹腔注射BALB/c小鼠,其中三株(Ⅱ_1B_7、Ⅱ_4B_4和Ⅱ_1H_7)获得腹水,纯化抗体并对其特性进行鉴定,三株单抗均可与BudR、IudR反应,而不与CdR、TdR及FudR反应,具有较高的特异性。
Immunization of BALB/c mice with bromodeoxyuridine (BudR) and bovine r-globulin (BGG) as antigens (BudR-BGG) using iodine reduction method, spleen cells and mouse myeloma cells (653) After fusion and screening and cloning by radioimmunoassay (RIA), four positive hybridomas II_1B_7, II_4B_4, I_3F_(11) and II_1H_7 were obtained. Hybridoma cells were injected intraperitoneally with BALB/c mice, three of them (II_1B_7, II_4B_4, and II_1H_7) obtained ascites. Antibodies were purified and characterized. Three monoclonal antibodies reacted with BudR and IudR but not with CdR. , TdR and FudR reaction, with high specificity.
