目的建立结直肠癌组织微阵列,探讨上皮钙黏附素和β链接素的表达在结直肠癌发生、发展过程中的作用。方法应用微阵列制作机制作结直肠正常黏膜腺瘤腺癌微阵列,用免疫组织化学法检测组织微阵列22例结直肠腺瘤和结直肠癌63例中上皮钙黏附素和β链接素的表达。结果制作了85例患者的结直肠腺瘤腺癌组织微阵列;原发各层腺癌组织及其转移灶中出现不同程度的β链接素核阳性表达,与正常黏膜相比远处转移灶除外,β链接素细胞核内表达增加,差异有显著性意义(P<0.01);上皮钙黏附素和β链接素膜表达在结直肠癌的发生、发展过程中呈下降趋势,肌层内癌、浆膜层内癌和淋巴结转移癌灶与正常黏膜、腺瘤及黏膜、黏膜下癌相比,β链接素膜表达减少(P<0.01),差异有显著性意义;正常黏膜与各癌灶,淋巴结转移灶与原发各层癌组织之间上皮钙黏附素的膜表达差异有显著性意义(P<0.05)。结论结直肠腺瘤腺癌组织微阵列的建立使得结直肠癌相关基因或蛋白的筛选工作快捷、简便,黏附分子上皮钙黏附素和β链接素的表达水平及表达部位异常与结直肠癌的发生、浸润转移有密切的关系。
Objective To establish a tissue microarray of colorectal cancer to investigate the role of epithelial cadherin and β-catenin expression in the development and progression of colorectal cancer. METHODS: Microarrays were used to prepare colorectal adenocarcinoma normal colorectal adenocarcinoma microarrays. Immunohistochemical method was used to detect the expression of E-cadherin and β-catenin in tissue microarray in 22 cases of colorectal adenoma and 63 cases of colorectal cancer. . Results The colorectal adenoma adenocarcinoma tissue microarray was prepared in 85 patients; the positive expression of β-linked nucleus was different in the primary layers of adenocarcinoma tissue and its metastases, except for the distant metastases compared with the normal mucosa. The expression of β-catenin was increased in nuclei, and the difference was significant (P<0.01). The expression of E-cadherin and β-catenin in the progression and progression of colorectal cancer showed a downward trend. Compared with normal mucosa, adenoma and mucosa, and submucosal cancer, the expression of β-catenin membrane was decreased (P<0.01) in intratumoral cancer and lymph node metastasis (P<0.01). There was significant difference between normal mucosa and each foci and lymph nodes. There was a significant difference in the expression of epithelial cadherin between the metastases and the primary cancer tissues (P<0.05). Conclusion The establishment of colorectal adenocarcinoma adenocarcinoma tissue microarray can make the screening of colorectal cancer-related genes or proteins work quickly and simply. The expression levels and abnormal expression of adhesion molecules E-cadherin and β-catenin are associated with the occurrence of colorectal cancer. There is a close relationship between infiltration and migration.
