目的通过分析PTEN、E-cd和MMP-9在人脑胶质瘤中的表达,为胶质瘤的靶分子治疗提供依据。方法采用免疫组织化学方法检测50例人脑胶质瘤和11例正常脑组织中的PTEN、E-cd蛋白和MMP-9的表达。分析人脑胶质瘤组织学分级之间的PTEN、E-cd蛋白和MMP-9的表达关系及胶质瘤与正常脑组织之间的PTEN、E-cd蛋白和MMP-9的表达关系,并探讨其临床意义。结果PTEN、E-cd在正常脑组织中的阳性表达为100%,PTEN在Ⅰ~Ⅱ级和Ⅲ~Ⅳ级胶质瘤中阳性表达率分别为75%和38%,组织学分级之间有显著差异(P<0.05),与对照组之间差异有统计学意义(P<0.05);E-cd在Ⅰ~Ⅱ级和Ⅲ~Ⅳ级胶质瘤中阳性表达率分别为42%和23%,组织学分级之间差异无统计学意义(P>0.05),与对照组之间差异有统计学意义(P<0.05);MMP-9在对照组阳性表达率为14%,在脑胶质瘤Ⅰ~Ⅱ级为33%,Ⅲ~Ⅳ级为62%,两者比较差异有统计学意义(P<0.05);对照组与神经胶质瘤组相比,MMP-9表达差异有统计学意义(P<0.05)。结论PTEN、E-cd、MMP-9在神经胶质瘤的生物学行为中起重要作用,且可能与其侵袭性有关。
Objective To analyze the expression of PTEN, E-cd and MMP-9 in human glioma and provide basis for target therapy of glioma. Methods Immunohistochemistry was used to detect the expression of PTEN, E-cd and MMP-9 in 50 human gliomas and 11 normal brain tissues. The relationship between the expression of PTEN, E-cd and MMP-9 in human glioma histological grading and the expression of PTEN, E-cd and MMP-9 between glioma and normal brain tissue were analyzed. And explore its clinical significance. Results The positive expression of PTEN and E-cd in normal brain tissue was 100%. The positive expression rates of PTEN in grade Ⅰ ~ Ⅱ and grade Ⅲ ~ Ⅳ gliomas were 75% and 38%, respectively (P <0.05), and the difference between the two groups was statistically significant (P <0.05). The positive expression rates of E-cd in grade Ⅰ ~ Ⅱ and grade Ⅲ ~ Ⅳ gliomas were 42% and 23% %, Histological grade was no significant difference (P> 0.05), and the difference between the control group was statistically significant (P <0.05); MMP-9 in the control group positive expression rate was 14% Compared with glioma group, the expression of MMP-9 in the control group was statistically significant (P <0.05), the difference between the two groups was statistically significant (P <0.05) Significance (P <0.05). Conclusion PTEN, E-cd and MMP-9 play an important role in the biological behavior of glioma and may be related to their invasiveness.
