目的研究大肠癌VEGF-C和VEGFR-3 mRNA的表达与大肠癌淋巴管生成的关系。方法采用RT-PCR检测32例新鲜大肠癌及相应正常组织中VEGF-C和VEGFR-3 mRNA表达,免疫组化染色淋巴管内皮特异标记物podoplanin计数淋巴管密度,以血管标记物CD34染色做对比。结果大肠癌VEGF-C及其受体VEGFR-3 mRNA水平显著高于正常组织,且二者表达具有显著正相关性。大肠癌VEGF-C mRNA高表达组癌周淋巴管密度显著高于低表达组。伴有淋巴管受累或淋巴结转移组VEGF-C mRNA显著高于无淋巴管受累或淋巴结转移组。结论VEGF-C/VEGFR-3信号转导途径可能对大肠癌癌周淋巴管生成具有重要作用,并可能进而促进癌组织淋巴结转移。
Objective To study the relationship between the expression of VEGF-C and VEGFR-3 mRNA and the lymphangiogenesis in colorectal cancer. Methods The expressions of VEGF-C and VEGFR-3 mRNA were detected by RT-PCR in 32 cases of fresh colorectal carcinoma and their corresponding normal tissues. The lymphatic vessel density was counted by immunohistochemical staining of podoplanin, a specific marker of lymphatic endothelial cells, and compared with vascular marker CD34 staining . Results The expression of VEGF-C and its receptor VEGFR-3 mRNA in colorectal cancer was significantly higher than that in normal tissue, and the expression of VEGF-C and VEGFR-3 was significantly positively correlated. The expression of VEGF-C mRNA in colorectal cancer was significantly higher than that in the low expression group. The expression of VEGF-C mRNA in lymphatic vessel involvement or lymph node metastasis was significantly higher than that in non-lymphatic vessel involvement or lymph node metastasis. Conclusion The VEGF-C / VEGFR-3 signal transduction pathway may play an important role in lymphangiogenesis in colorectal cancer and may further promote lymph node metastasis in colorectal cancer.
