The authors identified a solvent system for thin-layer chromatography that allows rapid isolation of benzylpenicillin (A) and its degradation products. Degradation substances common in aqueous solution were selected as the separation objects: Benzyl Penicilloic acid (B); benzyl Penicillic acid (C); benzyl Penicillenic acid (D) and α-methyl penicilloates (E). The samples were dissolved in water, spotted 5 micrograms on a silica gel plate (20 × 20 cm), the spreading distance was 15 cm, and the spreading groove was 7 × 28 × 22 cm. The filter paper was used to saturate the solvent in the groove wall. In the case of apparent layer, spray the reagent with potassium ferricyanide first, then place the chromatographic plate in the iodine vapor. The results showed that benzyl-penicillin was mixed with various degradation products by acetone-chloroform-acetic acid (50:45:5)
