目的观察疏肝饮对内脏高敏感性大鼠结肠组织中辣椒素受体(TRPV1)和降钙素基因相关肽(CGRP)表达的影响。方法将48只Wistar雄性大鼠随机分为正常组、模型组、疏肝饮组和得舒特组,采用慢性避水应激法造模10天。从造模第4天起,各给药组大鼠于造模前1小时分别给予相应的药物灌胃,连续7天,第10天造模结束后,采用结直肠气囊扩张引起的腹壁撤回反射来评估内脏敏感性;取出结肠组织,采用免疫组化法分别检测TRPV1和CGRP含量。结果与正常组比较,模型组的压力阈值降低(P<0.01),说明模型组大鼠存在内脏敏感性增高;与模型组比较,疏肝饮组和得舒特组的压力阈值明显增高(P<0.05)。与正常组比较,模型组结肠中TRPV1的表达及CGRP的含量显著增加,差异有统计学意义(P<0.01);与模型组比较,疏肝饮组和得舒特组大鼠结肠组织中TRPV1和CGRP含量均明显减少,差异有统计学意义(P<0.01)。结论疏肝饮通过调节结肠组织中TRPV1和CGRP的表达,从而降低内脏高敏感性。
Objective To observe the effect of Shugan decoction on the expression of capsaicin receptor (TRPV1) and calcitonin gene related peptide (CGRP) in the colon of visceral hypersensitive rats. Methods Forty-eight Wistar male rats were randomly divided into normal group, model group, Shugan Yin group and Decutamide group. The model was established by chronic water stress for 10 days. From the 4th day of modeling, the rats in each administration group were administered with the corresponding drugs one hour before the model making for 7 consecutive days. After the model was finished on the 10th day, the abdominal wall retraction reflex caused by the expansion of colorectal balloon To assess visceral sensitivity; remove the colon tissue, immunohistochemistry were detected TRPV1 and CGRP levels. Results Compared with the normal group, the pressure threshold of the model group decreased (P <0.01), which indicated that the visceral sensitivity increased in the model group. Compared with the model group, the threshold pressure of Shugan Decoction and Dictax group increased significantly (P <0.01) <0.05). Compared with the normal group, the expression of TRPV1 and the content of CGRP in the model group were significantly increased (P <0.01). Compared with the model group, the expression of TRPV1 in the Shugan Yin group and the DST group And CGRP levels were significantly reduced, the difference was statistically significant (P <0.01). Conclusion Shugan Yin can reduce visceral hypersensitivity by regulating the expression of TRPV1 and CGRP in colon tissue.
