目的:探讨Kif2a在人上皮卵巢癌细胞中的表达及其沉默对癌细胞增殖、侵袭及凋亡的影响。方法:选取人上皮卵巢癌细胞株(OVCAR-3,SKOV-3)及人正常卵巢上皮细胞株(IOSE-80)进行常规培养;采用荧光定量PCR(q-PCR)和Western blot法检测各细胞中Kif2a基因及蛋白的表达量;以siRNA干扰法对癌细胞中的Kif2a基因进行沉默,沉默效果以q-PCR及Western blot法验证;MTT法检测细胞增殖;Transwell实验检测细胞侵袭能力;细胞凋亡率以流式细胞术进行检测;凋亡相关蛋白active-caspase 3及Bcl-2表达以Western blot法检测。结果:Kif2a在人上皮卵巢癌细胞中的表达均显著高于人正常卵巢上皮细胞。Kif2a基因沉默后,OVCAR-3和SKOV-3细胞增殖及侵袭能力均显著下降;细胞凋亡率显著增加,并伴随促凋亡蛋白activecaspase 3表达显著上升及抑凋亡蛋白Bcl-2表达显著下降。结论:Kif2a在人上皮卵巢癌细胞中表达显著上升,其沉默能够抑制上皮卵巢癌细胞的增殖和侵袭,并诱导细胞凋亡。
Objective: To investigate the expression of Kif2a in human epithelial ovarian cancer cells and its effect on the proliferation, invasion and apoptosis of cancer cells. Methods: Human ovarian epithelial ovarian cancer cell line (OVCAR-3, SKOV-3) and human normal epithelial ovarian epithelial cell line (IOSE-80) were selected for routine culture. The cells were detected by qRT-PCR and Western blot In Kif2a gene and protein expression levels; Kif2a gene silencing of cancer cells by siRNA interference silencing effect by q-PCR and Western blot validation; MTT assay of cell proliferation; Transwell assay of cell invasiveness; apoptosis The apoptosis rate was detected by flow cytometry. The expressions of active-caspase 3 and Bcl-2 were detected by Western blot. Results: The expression of Kif2a in human epithelial ovarian cancer cells was significantly higher than that in normal human ovarian epithelial cells. After Kif2a gene silencing, the proliferation and invasion ability of OVCAR-3 and SKOV-3 cells were significantly decreased; the apoptosis rate was significantly increased with the increase of activecaspase 3 expression and the decrease of Bcl-2 expression . Conclusion: Kif2a expression in human epithelial ovarian cancer cells increased significantly, and its silencing can inhibit epithelial ovarian cancer cell proliferation and invasion, and induce apoptosis.
