目的利用DNA条形码二级结构鉴定方法,快速、准确的鉴别花椒及其混伪品。方法采用改良的DNA试剂盒提取花椒及其混伪品基因组DNA,对ITS2序列扩增和测序。用Neighbor-joining(NJ)系统聚类结果与ITS2序列二级结构相比对的方法进行鉴别分析。结果花椒、竹叶花椒与野花椒分别有2种类型ITS2二级结构,青椒有1种类型ITS2二级结构;中药花椒二基源物种与竹叶花椒、野花椒的二级结构不同,可明显区分。结论 ITS2序列二级结构作为DNA条形码辅助分析手段可准确有效鉴别中药花椒及其混伪品。
Objective To identify the pepper and its counterfeit products quickly and accurately by using the DNA secondary structure identification method. Methods Genomic DNA of Zanthoxylum bungeanum and its adulterated product was extracted by a modified DNA kit, and the ITS2 sequence was amplified and sequenced. The Neighbor-joining (NJ) cluster analysis was compared with the secondary structure of ITS2 sequence. Results There were two types of ITS2 secondary structure in Zanthoxylum bungeanum, Zanthoxylum bungeanum and Zanthoxylum bungeanum, respectively, and one type of ITS2 secondary structure in Zanthoxylum bungeanum. Conclusion The secondary structure of ITS2 sequence can be used as an aid to DNA barcode analysis to identify Chinese traditional medicine pepper and its counterfeit products accurately and effectively.
