目的制备负载热休克诱导凋亡的脑胶质瘤细胞的树突状细胞(DC),探讨其激发T细胞的作用。方法诱导正常人外周血来源未成熟DC,负载热休克诱导凋亡的脑胶质瘤细胞的树突状细胞,通过双荧光标记法流式检测DC表型、MTT法测定CTL活性。结果负载热休克脑胶质瘤细胞的DC具有良好的激活CTL作用及对胶质瘤的杀伤效应也显著高于负载冻融裂解脑胶质瘤细胞制备的DC,差异有统计学意义(P<0.05),而且杀伤活性随着效靶比而增加。结论负载热休克脑胶质瘤细胞的树突状细胞是一种具有潜在临床应用价值的肿瘤疫苗。
Objective To prepare dendritic cells (DCs) loaded with heat shock-induced apoptosis of glioma cells and to explore their role in stimulating T cells. Methods DCs from normal human peripheral blood were loaded with dendritic cells loaded with heat shock to induce apoptosis of DCs. The DC phenotypes were detected by flow cytometry. The CTL activity was measured by MTT assay. Results The DCs loaded with heat shock glioma cells had a good activated CTL effect and the killing effect on glioma was also significantly higher than those loaded with lytic-thawing lysed glioma cells, the difference was statistically significant (P < 0.05), and the killing activity increased with the target ratio. Conclusion Dendritic cells loaded with heat-shocked glioma cells are potentially valuable tumor vaccines.
