以2个黄瓜品种“金谷六号”和“吉列168”的种子为试材,以子叶和下胚轴作为外植体进行离体培养,研究了不同培养基和激素浓度及配比对黄瓜愈伤组织诱导、不定芽分化的影响。结果表明:2种不同基因型的黄瓜均表现出较强的愈伤组织分化能力,下胚轴要优于子叶,不同激素配比组合对愈伤组织的形态影响较大,AgNO_3能有效抑制愈伤组织的玻璃化。2种黄瓜以子叶为外植体的最佳激素配比均为MS+0.5mg·L~(-1) 6-BA+0.10mg·L~(-1) IAA+1.0mg·L~(-1)AgNO_3;2种黄瓜以下胚轴为外植体的最佳激素配比均为MS+2.0mg·L~(-1) 6-BA+0.10mg·L~(-1)IAA+1.0mg·L~(-1) AgNO_3。该试验条件下不定芽的分化没有形成。
