目的:探讨结直肠腺瘤与非肿瘤性息肉中转化生长因子-β1(TGF-β1)的表达差异及其与细胞增殖的关系。方法:采用免疫组化S-P法检测36例结直肠腺瘤、36例非肿瘤性息肉、13例腺癌和11例正常黏膜组织中TGF-β1和Ki-67的表达;采用原位杂交技术检测TGF-β1mRNA。结果:TGF-β1蛋白及其mRNA和Ki-67在结直肠腺瘤组的表达均高于非肿瘤性息肉组(P<0·05);结直肠腺瘤组PI值与TGF-β1的表达强度呈显著正相关(rs=0·557,P<0·01)。结论:腺瘤组织中TGF-β1与细胞增殖活性密切相关,TGF-β1在腺瘤的高表达和非肿瘤性息肉的低表达可能是二者恶变趋势差异的重要分子机理之一。
Objective: To investigate the expression of transforming growth factor-β1 (TGF-β1) in colorectal adenomas and non-tumor polyps and its relationship with cell proliferation. Methods: Immunohistochemical SP method was used to detect the expression of TGF-β1 and Ki-67 in 36 cases of colorectal adenoma, 36 cases of non-tumor polyp, 13 cases of adenocarcinoma and 11 cases of normal mucosa. The in situ hybridization TGF-β1 mRNA. Results: The expression of TGF-β1 protein and its mRNA and Ki-67 in colorectal adenoma group were significantly higher than those in non-tumor polyp group (P <0.05). The PI value and the expression of TGF-β1 in colorectal adenoma group There was a significant positive correlation between intensity (rs = 0.557, P <0.01). CONCLUSION: TGF-β1 is closely related to cell proliferation in adenomas. The high expression of TGF-β1 in adenomas and the low expression of non-tumorous polyps may be one of the important molecular mechanisms of the malignant transformation.
