在昆明(KM)小鼠标准化研究中,采集武汉生物制品研究所、上海第一生化制药厂普通级KM小鼠的精子并冷冻运输,冻融精子与中科院上海实验动物中心KM雌鼠的超排卵体外受精(IVF)得到杂交一代2细胞胚胎,用中科院动物中心KM鼠冻融精子对照。无菌条件下2细胞胚胎移植入SPF级假孕鼠,受体饲养于独立通气笼具(IVC)中。平均产仔率为51.9%,离乳率100%。三种群12周龄KM雄鼠精子冷冻复苏率,IVF率,移植成绩均无显著差异(P>0.05)。仔鼠经微生物、寄生虫学检测达SPF标准。结果表明,利用冷冻精子,通过IVF、胚胎移植能够方便快速的建立种群间杂交一代,并实现生物净化。
In Kunming (KM) mouse standardization study, the sperm from the Wuhan Institute of Biological Products, Shanghai First Biochemical Pharmaceutical Factory of ordinary KM mice were collected and frozen for transport. The frozen-thawed sperm and the superovulation of KM female mice in Shanghai Experimental Animal Center of Chinese Academy of Sciences In vitro fertilization (IVF) to get a generation of hybrid 2-cell embryos, using the Chinese Academy of Sciences Animal Center mouse frozen-thawed sperm control. Two-cell embryos were transplanted into SPF pseudo-pregnant mice under aseptic conditions and the recipients were housed in independent ventilation cages (IVC). The average litter size was 51.9%, the rate of 100% milk. There was no significant difference in sperm cryogenic resuscitation rate, IVF rate and transplantation among three groups of 12-week-old male and female KM rats (P> 0.05). Offspring by microorganisms, parasites test up to SPF standards. The results show that the use of frozen sperm, IVF, embryo transfer can quickly and easily establish interbreeding hybrids, and to achieve biological purification.
