染色体片段置换系(chromosome segment substitution lines,CSSL)基因组内只含有一个来自供体亲本的染色体片段,而基因组的其余部分与轮回亲本相同,并能把QTL拆分为单个的孟德尔因子,因此CSSL是进行基因组分析,特别是QTL分析的理想材料.本研究以陆地棉遗传标准系TM-1为受体亲本,海岛棉海7124为供体亲本在BC5S1代借助分子标记辅助选择(marker-assisted selection,MAS)培育了一套染色体片段置换系.这套置换系由330个株系构成;1~4个不同的株系携带相同的染色体片段,置换片段的平均长度为10.9cM,总长度为5271.9cM,是棉花基因组总长度3514.6cM的1.5倍.每个株系内被替换的染色体片段长度不一,最短为3.5cM,最长为23.2cM.由于还有些区段缺失供体亲本的染色体片段,所以这个替换系群体没有完全覆盖棉花基因组.
The chromosome segment substitution lines (CSSL) genome contains only one chromosome segment from the donor’s parent, while the rest of the genome is identical to the recurrent parent and splits the QTL into a single Mendelian factor, thus CSSL Which is the ideal material for genome analysis, especially for QTL analysis.In this study, the genetic cotton TM-1 was used as the parent and the cotton seedlings of Qingdao 7121 as the donor were selected by marker-assisted selection in BC5S1 , MAS) developed a set of chromosome segment replacement lines consisting of 330 lines; 1 to 4 different lines carrying the same chromosomal segment, the average length of the replacement segment was 10.9 cM and the total length was 5271.9 cM, which is 1.5 times of the total genome length of 3514.6cM in cotton.The length of chromosomal fragments substituted in each line varied from 3.5cM to 23.2cM in length.The chromosome segments of donor’s parents were deleted in some segments , So this population of substitution lines did not completely cover the cotton genome.
